What is Trace File/Chomatogram
After sequencing of a PCR product, the sequencing machine operator will provide a trace file or chromatogram. "Trace file provides the nucleotide base order(ATCGTCGAACGT) of the DNA."
Every sequence should have two trace files. First one is for Forward sequence(sequenced with forward primer) and Second one is for complementery Reverse sequence (sequenced with Reverse primer).
Every nucleotide is defined with a particular color code worldwide like A(green), T(red), G(black) and C(Blue). The actual nucleotide will be identified initially by observing the peak in a chromatogram and comparing with the NCBI sequences in Genbank by Blasting and then by comparing between Forward and Reverse sequences.
